چکیده
Aspergillus strains obtained from IMTECH Chandigarh and from Department of Biotechnology, CDLU, Sirsa were screened for their ability to produce amylase by starch agar plate method. Among the five strains, A. niger MTCC-104 showed highest clearing zone on starch agar plates as well as amylase activity in solid state fermentation. Different substrates like wheat bran, rice bran, soybean meal and black gram bran were screened for enzyme production. Rice bran was found to be the best substrate for the enzyme production. This rice bran supplemented with aqueous salt solution was used for further studies of amylase production and characterization. The enzyme amylase was found to be thermo-stable and active at wide range of pH. Enzyme showed maximum activity at 70C. J.S. DUHAN Department of Biotechnology, Ch. Devi Lal University, Sirsa-125055

Amylase production from Aspergillus oryzae LS1 by solid-state fermentation and its use for the hydrolysis of wheat flour
Nine Aspergillus and three of Trichoderma strains were grown on wheat bran (WB) medium under solid state fermentation (SSF) for amylase production. Aspergillus oryzae LS1 produced the highest level of the enzyme. The thermal stability profile of its crude enzyme revealed the half-life time of more than 2 h at 50 and 60ºC. The enzyme production was affected by strain type, incubation periods, le...
متن کاملOptimization of Amylase and Protease Production from Aspergillus awamori in Single Bioreactor Through EVOP Factorial Design Technique
Evolutionary operation (EVOP) factorial design technique was explored in order to economically produce amylase and protease at their optimum level in a single bioreactor by modified solid-state fermentation. Maximum yields of amylase and protease were achieved, using wheat bran as a substrate by a highly potent, locally isolated strain of Aspergillus awamori: Nakazawa MTCC 6652. The strain had ...
متن کاملProduction and Characterization of "-Amylase from Aspergillus niger JGI 24 Isolated in Bangalore
Five fungal isolates were screened for the production of "-amylase using both solid-state and submerged fermentations. The best amylase producer among them, Aspergillus niger JGI 24, was selected for enzyme production by solid-state fermentation (SSF) on wheat bran. Different carbon and nitrogen supplements were used to enhance enzyme production and maximum amount of enzyme was obtained when SS...

Demand for microbial amylase production persists because of its immense importance in wide spectrum industries. The present work has been initiated with a goal of optimization of solid state fermentation condition for amylase using agroindustrial waste and microbial strain like B. amyloliquefaciens (MTCC 1270). In an aim to improve the productivity of amylase, fermentation has been carried out ...
متن کاملRaw Starch Degrading Amylase Production by Various Fungal Cultures Grown on Cassava Waste
The solid waste of sago industry using cassava was fermented by Aspergillus niger, Aspergillus terreus and Rhizopus stolonifer in solid state fermentation. Cassava waste contained 52 per cent starch and 2.9 per cent protein by dry weight. The amylase activity was maintained at a high level and the highest amylase activity was observed on the 8(th) day in R. stolonifer mediated fermentation. R. ...
